Semaglutide Reduces Body Weight Regardless of Menopause Status: STEP and OASIS 4 Post Hoc Analysis (144116)

Background

This post hoc analysis investigated the impact of semaglutide on anthropometric measures by menopause status among women with overweight or obesity in the STEP and OASIS 4 trials.

Methods

Female participants (pts) with body mass index ≥30 kg/m2, or ≥27 kg/m2 with ≥1 obesity-related complication who received once-weekly subcutaneous semaglutide 2.4 mg for 68 weeks (wks) in STEP 1, 3, 4, 5, 8, and 9, or once-daily oral semaglutide 25 mg for 64 wks in OASIS 4, were stratified by menopause status. Endpoints were changes in body weight (BW), waist circumference (WC), and waist-to-height ratio (WHtR).

Results

Overall, 2151 STEP pts (pre, n=876; peri, n=414; post, n=861), and 151 OASIS 4 pts (n=68, n=33, n=50, respectively) were included. For the STEP trials, median (interquartile range) % changes in BW from BL to wk 68 for pre, peri, and post groups were: −18.0% (−26.3, −9.9), −17.7% (−23.7, −10.8), and −16.4% (−22.5, −9.9), respectively; for OASIS 4, changes in BW from BL to wk 64 were: −18.1% (−26.5, −7.2), −15.6% (−23.5, −10.5), and −15.6% (−19.8, −6.0), respectively. Similar proportions of STEP pts achieved ≥10% weight loss (WL) across the pre, peri, and post groups (74.7%, 78.7%, and 74.8%, respectively). Mean (standard deviation) change from BL in WC (cm) in the pre, peri, and post groups were: −18.5 (12.0), −17.7 (10.3), and −15.9 (9.1) (STEP); and −17.7 (13.5), −15.3 (8.6), and −13.5 (10.3) (OASIS 4); respectively. The proportion of pts in the low-risk WHtR category increased from BL in the pre, peri, and post groups: 17.1%, 14.2%, and 9.1% (STEP); and 19.0%, 9.7%, and 8.3% (OASIS 4); respectively.

Conclusions

Semaglutide was associated with WL and improvements in anthropometric parameters in women with overweight or obesity without diabetes, regardless of menopause status.
